An ATP-synthesizing system in lettuce (Lactuca sativa L.) seed powder is demonstrated. The system is AMP- and PEP- (phosphoenolpyruvate) dependent and has a pH optimum at 6.4. There is a substrate inhibition by AMP at concentrations above 2 mM. PEP has an apparent Km value of 8 mM. The system is stimulated by NADH2. The accumulation of ATP is linear up to about 40 min, followed by a sharp decrease between 1 and 4 h of incubation. This decrease is shown to be a result of: (a) loss in the ability to synthesize ATP during the incubation time; (b) utilization of ATP for protein and nucleic acid synthesis and (c) possible “aging” processes and degeneration of cells during incubation, resulting in hydrolytic activities.
