Pendant la formation du spermatophore de Penaeus kerathurus, deux couches, I et II, entourent la masse des spermatozoïdes dans un seul conduit de la portion ascendante du canal déférent. Elles forment un corps cylindrique principal, alors que deux autres couches, III et IV, simultanément déposées dans un conduit annexe, constituent l'aile accessoire. Une connexion préliminaire a lieu entre ces deux parties par la couche V, qui est déposée à l'origine autour de l'aile, mais qui, ensuite, entoure incomplètement le corps du spermatophore.
