Axenic culturing of Aphelenchoides rutgersi in an aqueous diet containing nematicidal and nematistatic chemicals appears to be a useful technique for toxicological and mode of action studies. Reproduction was significantly retarded or totally stopped at 10 ppm (w/v) in a 7-day test that is applicable for screening chemicals potentially useful for controlling nematodes. During test periods lasting up to 21 days, reproduction was unaffected when 1 ppm Nemafos, Aldicarb or DuPont 1410 were added to the diet of this nematode; slightly retarded when Furadan was used; but severely retarded when Dasanit or Nemacur was included. However, stimulation of reproduction occurred when Aldicarb, Nemacur, or DuPont 1410 were incorporated at 0.1 ppm. The effects of these chemicals on total numbers, number of eggs, percent adult females, and percent mortality were determined.
